Abstract
Cetuximab, an anti-epidermal growth factor receptor monoclonal antibody, has been shown to increase the median survival of colon cancer patients. To establish if egg serum albumin nanoparticles (ESA-NPs), a drug carrier, could be a factor that renders colon cancer cells to be more susceptible to chemotherapy’s cellular cytotoxicity of cetuximab. In the present study, we prepared ESA-NPs and cetuximab-loaded albumin nanoparticles (CET-ANPs) using a desolvation method, while ESA and the nanoparticles were characterized by X-ray diffraction (XRD)- analysis to determine the shapes of the particles of substances and by the transmission electron microscope (TEM) to determine the nanoparticles’ shapes and sizes. Using MTT assay for cellular cytotoxicity, cetuximab- and CET-ANPs- mediated significant cytotoxicity against the colon cancer Caco-2 cells after 48 h. We observed that the colon cancer cells treated with cetuximab were killed more efficiently by CET-ANPs after 48 hrs. Interestingly, the treatment of target cells with different concentrations of cetuximab and ESA-NPs achieved higher efficiency in treatment. In conclusion, the present results indicate and predict that loading Cetuximab with ESA-NPs showed more efficiency.
