Abstract | ||||
Background/aims. Blood glucose continuously fluctuates in healthy individuals. This study compared the risk of acute glucose fluctuation and constant hyperglycemia in the aorta of non-diabetic animal models. The possible therapeutic effect of granulocyte-colony stimulating factor (G-CSF) was investigated. Methods. Thirty male Wistar rats were divided into (Gr-1) control, (Gr-2) constant hyperglycemia (Gr-2), (Gr-3) acute glucose fluctuating induced by injection of 1 ml 50% glucose solution intraperitoneal every 4 hours for 4 days, (Gr-4) Recovery group: like (Gr-3) but did not receive anything for 7 days then sacrificed and (Gr-5) treated group. After induction of glucose fluctuation, rats were injected with 200 microgram/kg/day of diluted G-CSF (Neupogen®) subcutaneously for 5 days. Blood levels of glucose, Insulin and C-peptide were measured. The aorta was examined histologically by (Hematoxylin & Eosin, Masson and Orcein), immunohistochemically by Caspase-3 and VEGF markers and by electron microscope. Results: Glucose returned to normal in Gr-4 and Gr-5. Insulin levels decreased in Gr-2, Gr-3, Gr-4 and Gr-5. C-peptide decreased in Gr-2 and Gr-3. Gr-4 and Gr-5 showed no significant decrease in C-peptide. Gr-3 showed endothelium damage, destructed mitochondria, an increase in collagen and disrupted elastic lamellae. Gr-2 showed less damage than Gr3. Gr-4 showed limited structural improvement. Gr-5 showed normal cell proliferation .Gr-3 showed an increase in the amount of collagen deposition while Gr-4 and Gr-5 showed significant decrease in collagen. The elastic lamellae in Gr-2 and Gr-3 were disrupted and irregular, while in Gr-4 and Gr-5 were regularly organized . Caspase-3 expression was weak in the endothelium of Gr-2 and Gr-3 and strong in smooth muscles while Gr-4 and Gr-5 showed decreased expression. VEGF expression was weak in Gr-1, strong in Gr-3, strong in endothelium weak in smooth muscle in Gr-2, and weak in Gr-4 and Gr-5. Conclusion: AGF causes more aortic damage. G-CSF ameliorate these changes. | ||||
