Abstract | ||||
The aim is to evaluate the role of P21 and SOX9 genes methylation in de novo hepatocellular carcinoma (HCC) after direct-acting antivirals (DAAs) therapy for hepatitis C virus (HCV). The study included 74 cases, and 20 age- and sex-matched control divided into 5 groups: Group 1: patients with liver cirrhosis who developed de novo HCC after DAAs with viral cure. Group 2: liver cirrhosis with de novo HCC without receiving DAAs. Group 3: liver cirrhosis treated with DAAs with viral cure. Group 4: liver cirrhosis not treated by DAAs. Group 5: healthy controls. Data was recorded and DNA methylation of P21 and SOX9 genes were measured. The mean P21 and SOX9 genes methylation percentages were significantly higher in the cases and showed the highest values in group 1. At cut-off of ≥ 93.6%, P21 methylation showed 95.7% sensitivity, 72.3% specificity, AUC 0.836 and p=0.000. P21 methylation was significantly correlated with HCC (r= 0.584, p= 0.000) while SOX9 methylation was correlated with DAAs (r= 0.411, p= 0.000). P21 gene methylation can be an epigenetic biomarker in de novo HCC after DAAs. SOX9 gene methylation could be induced by DAAs but not correlated to HCC. | ||||
