Abstract | ||||
Background: Urinary tract infections (UTIs) are among the most prevalent bacterial infections in the world. P.mirabilis's ability resistance to a number of antibiotic classes. to evaluate the phenotypic and genotypic characteristics of P.mirabilis isolates from patients with UTIs in order to identify resistance genes and in addition to knowledge about DNA sequence. Aim: The purpose of this study was to identify, categorize, and isolate P.mirabilis. bacteria into phylogroups and strains to determine the relationships between isolates taken from clinical samples in in Najaf Governorate. and Detection of resistance gene of P.mirabilis encoded by different genes by using PCR technique like bla oxa, bla ctx, bla shv, bla tem as well as 16S RNA .also Investigate the antibiotic resistance patterns for all isolates and Finally, study phylogenetic tree Methods: Samples collected from Najaf at a rate of 325 samples were subjected to culture, isolation and identification by biochemical tests , vitek2 compact system , PCR technique finally DNA sequence. Results: This work aims to isolate and identify P.mirabilis. responsible for various clinical infections in individuals. This study also aimed to detect bacterial P.mirabilis with antibiotic resistance. Recovered P.mirabilis showed resistance to many antibiotics out of 17 examined antibiotics based on antibiotic susceptibility test. Resistance to many drugs 20 All of the resistance genes (bla tem, bla ctx, bla shv, and bla oxa) are present in varying amounts in P. mirabilis isolates. Of the 20 isolates, 17 had the presence of genes that encode bla tem, with an amplification product of 766 bp; the other isolates were negative.11positive isolates with an amplification product of 536 bp were obtained using bla ctx, while the remaining isolates were negative. 8 isolates out of 20 had the presence of genes encoding bla shv, with an amplification product of 827 bp; the other isolates tested negative. However, bla oxa alone produces unfavorable outcomes. The genomic DNA of the primers of the diagnostic gene16Sr RNA was analyzed after taking 20 micrograms/ microliter of the amplified DNA of 20 of the studied isolates,(10)isolates from( P.mirabilis).After analyzing the results in the BLAST program available on the global website of the American gene bank National Center for Biotechnology Information (NCBI), it was found that the matching rate is very high with (P.mirabilis),so (8) isolates were registered in the bank and were given accession numbers ,all isolates registered for the first in Iraq .Conclusion: P.mirabilis infections are more common in females and show high resistance to several antibiotics, except meropenem. Genetic analysis highlights its diversity, resistance genes (bla tem, bla ctx, bla shv), and close global relations, marking a key milestone in Iraq's bacterial research. | ||||
