Abstract | ||
Background: Integrons are transposable elements found in bacteria, and they have clearly demonstrated their role in antibiotic resistance. Objective: This study aimed at gene expression to find alternatives derivative of antibiotic treatment for drug resistance of Acinetobacter baumannii. Methods: In the present study, mixed antibiotic meropenem with titanium dioxide (TiO2) NPs prepared by laser ablation in liquids was used. The diagnosis and antibiotic resistance pattern of bacteria are done by the bacteriological and biochemical test by DL-120E and devices ID and AST Vitek. Gene detection of integron Class1,2,3 by qPCR. Antimicrobial activity was determine by disc well diffusion agar and minimal inhibitory concentration (MIC). The MIC was used to determine the concentration for Meropenem because was choised to combination with NPs as a drug against A. baumannii total RNA purification, qPCR amplification, and data. That analysis and calculation of gene expression levels of one or more genes before and after nanoparticle use was detected depending on RNA concentration after conversion to cDNA. Results: Gene detection of bacteria carriers that have integrons, then exposure to mixed NPs, then isolation for more effective killing in the composite meropenem-TiO₂ nanoparticles against A. baumannii resistance, selected to study gene expression, showed regular and down-regular gene expression. Conclusion: our study was found anew treatment against MDR, XDR A. baumannii by study gene expression of integron that cause down- regulation and up- regulation gene create by MEM-TiO2 NPS and kill it. | ||
