| Abstract | ||
| Background: Pulmonary fibrosis is among the highly fatal respiratory diseases worldwide and the establishment of good experimental evaluation technique is essential to study the disease and evaluating possible treatment modalities. Many methods are used to evaluate spirometry like paramenters in animal model however classical method has its limitations such as the use of anesthesia and animal immobilization. Whole-body plethysmography (WBP) avoids these issues providing a non-invasive approach for the assessment of the respiratory function in conscious animals. Resveratrol recently introduced into the medical field for its antifibrotic,anti oxidant and anti-inflammatory. The aim of the present study was to investigate the ability of WBP as a tool to evaluate different ventilatory parameters which could differentiate respiratory disease pattern and alleviate the effectiveness of treatment. Methods: PF was induced in 20 Wistar male rats by single endotracheal injection of bleomycin, those were randomly divided into two groups (10 rats each): (untreated PF group) – Resvertrol treated PF group. Results: Induction of PF was associated with increased Respiratory rate and decrease Vt and MRV with normal FEV1% and normal flow rates. Treatment with Resveratrol showed marked improvement of the asssessed ventilatory parameters. conclusion: This protocol describes the procedures for the measurement of the breathing patterns in rats using the WBP method, the data analysis and results interpretation. | ||
