Abstract | ||||
Theaimofthecurrentstudywastoevaluatethe expressionofcaspase9asanapoptoticmarkerin7, 12- Dimethylbenz(a)anthracene (DMBA)inducedhamster buccalpouch (s)(HBP(s)) carcinoma.Materialand methods:Theexperimentalanimalsusedinthecurrentstudywerefifty-fivegoldenSyrianhamstersweighting 80-129g,andobtained fromthe animalhouse,CairoUniversity.The experimentalanimals weredividedinto two main groups:Group A (normalgroup) 5hamsters weretaken, not treated and served as control. Group B:Theremaininganimals(50hamsters)weretreatedwiththesameprocedures:HBPswerepaintedfor18 weekswithDMBA,3 timesaweek.Then,a directlesionalinjectionby differentkindsoftreatment:gold nanoparticles (AuNPs) (groupB2),AuNPs+laser (groupB3)cisplatin(groupB4),AuNPsconjugatedwith cisplatin and laser irradiation (group B5), were subsequentlyemployed. After that, various investigation: hematoxylinandeosin(H&E)stainandimmunohistochemicalstainingutilizingantibodyagainstcaspase9 weredone.Results:Grossobservationrevealedthatthereis avariationinthereductionofthedistribution andsizeof tumorsbetweenthe treatedgroups.Immunohistochemicalresultsrevealed thatcaspase 9expression hasawidevariability intheareapercentagedistributionthrough-outthetreatedgroups.Conclusions: AuNPsinconjunctionwithcisplatinandvisible laserirradiationhavepositiveeffectontumor ablation. Moreover, intra-tumoral application of AuNPs in conjunction with cisplatin and visible laser irradiation is considered as a promising therapeutic agent in the regression of HBP DMBA induced carcinoma. | ||||
