Abstract | ||||
ABSTRACT Objective: To investigate the toxic effects of doramectin (DRM); if any; on apoptosis, oxidative stress and fertility in female Wistar rats. Design: Descriptive study Animals: Thirty clinically healthy mature female Wister rats with estrus phase Procedures: Female rats with estrus phase were divided randomly into 3 equal groups. 1st group, female rats were injected with sesame oil subcutaneously as a single dose and kept as control group. 2nd group, female rats were injected subcutaneously with a single therapeutic dose of doramectin (200µg/kg B.wt). 3rd group, female rats were injected subcutaneously with double therapeutic dose of doramectin (400µg/kg B.wt). Blood samples were collected for measuring of the levels of serum E₂, FSH, LH and P4. Lipid peroxidase (LPO), Reduced glutathione (GSH), Catalase (CAT) activity were also measured. At the end of the experiment, ovaries, uteri and the anterior pituitary gland were collected for histopathological examination. Results: The results showed that there was significant decrease in all tested hormones except P4 (significant increase) in both treated groups, moreover, DRM caused significant alteration in the oxidative stress markers in both treated groups with some histopathological lesions in ovary, uterus and anterior pituitary gland. Conclusion and clinical relevance: Either single or double therapeutic dose of doramectin could induce several hormonal disturbances in female rats, also there were markable reduction in oxidative stress and antioxidant defense markers with some histopathological lesions were also observed. | ||||
