| Abstract | ||||
| Background and Objectives: Diabetic corneal morphological changes are frequent. Green tea extract (GTE) is a natural product which was proved to have antioxidant and anti-inflammatory effects. Adipose mesenchymal stem cells (AMSCs) are commonly used as cellular therapies. The current work pointed to studying and comparing the effect of therapy of AMSCs individually or combined with GTE in keratopathy complicating induced type1 diabetes (T1D) in adult male rats. Methods and Results: 34 male rats were randomly divided into: In Vitro Study Group: 2 rats for isolation, culture, and label of AMSCs. Group A: (Control Rats) 4 animals. Group B: (Diabetic Rats): 7 animals were injected intraperitoneal (IP) with 50 mg/kg streptozotocin (STZ). Group C: (GTE Group): 7 animals injected with streptozotocin, 4 weeks later GTE 50 mg/kg was given daily orally for another 4 weeks. Group D: (AMSCs Group): 7 animals injected IP with STZ, 4 weeks later, intravenous (IV) injection of 1x106 cells (AMSCs) was performed. Group E: (GTE and AMSCs Group): 7 animals injected with STZ, 4 weeks later, AMSCs were injected, and combined GTE therapy was performed. Animals of groups A, B, C, D, E were sacrificed after 8 weeks from start of experiment. Corneal dimensions, serological, biochemical, morphological, and quantitative studies were performed. Group B revealed inflammatory and degenerative changes in the various layers that regressed by various therapies. Conclusions: T1D induced corneal inflammatory and degenerative morphological changes. GTE and AMSCS proved amelioration of morphological and quantitative parameters, the amelioration was more remarkable in response to AMSCs therapy. Combined GTE and AMSCs therapy guaranteed the most remarkable effect, that can be related to activated trans-differentiation. | ||||
