| Objectives: Molecular investigation and phylogenetic analysis of PPMV-1 Design: Descriptive study Animals: Twenty-nine pigeons suffering from nervous signs and greenish diarrhoea were studied. Procedures: 19 out of 29 pigeons were euthanized and pooled organs (brain, spleen, lung and liver) were inoculated in ECE. Positive nine collected allantoic fluid for HA activity were examined by RT PCR using Primer sequence of 356 base pair of fusion protein gene [1] and then sequenced, analysed to determine the phylogenetic characterization of PPMV-1.Proteolytic cleavage sites (residues112-117) of F protein were identified as KRKKGF, KKKKAF. Results: Phylogenetic analysis based on f gene revealed that two isolates were aligned in genotype VIb/2 together with other PPMV-1 from Egypt, Iran and Croatia with accession No Pigeon/ Egypt/ MN610435 R7 Dak/2015 and Pigeon/ Egypt/ MN610436 R9 Dak/2018. The identity between our two strains were 96.62 % and between our strains and other Egyptian strains in phylogenetic tree were from 94.20 to 90.55. There was a multi basic amino acid (KRKKG) cleavage site at position 112 to 116 and a phenylalanine residue at position 117 for R9, but mono basic amino acids (KKKKA) for R7. Both velogenic and lentogenic strains of PPMV-1 can cause nervous manifestations. Conclusion and clinical relevance: The phylogenetic analysis indicates that both isolated velogenic and lentogenic strains of PPMV-1 can cause nervous manifestations. | ||||
