| Abstract | ||||
| This study aimed to examine the protective effect of white garlic powder (WGP) and black garlic powder (BGP) as supplemented to diet on hepato carcinoma induced by N-nitrosodiethylamine (NDEA) in male rats. Forty two male rats were randomized into 6 equal groups (n=7). Group 1 was kept as a normal control group ,while the other 5groups were intoxicated by a single intraperioneal dose (100 mg/kg) of NDEA in the 3rd week , followed by weekly subcutaneous injections with CCl4 till the end of the experiment(6 weeks) to induce hepatocarcinoma. Group 2 kept asa positive control group and groups 3,4, 5 and 6 were fed on diets containing WGP and BGPatlow (2.5%) and high (5%)concentrations, respectively .The changes of serum biochemical parameters, tumor biomarker sand liver histology were used to evaluate the protective effect of white and black garlic powder. Tissue lipid peroxidation, antioxidant defense capacity and content of 8-hydroxy-2-deoxyguanosine (8-OHdG)in liver DNA, were used as a markers of apoptosis, were measured to examine the mechanisms of protection .The results showed that WGP and BGP caused reductions of serum biochemical indices: aspartate aminotransferase, alanineaminotransferase, alkaline phosphatase and total bilirubin.Garlic powder ssuppressed tumor biomarkers: alpha fetoprotein(AFP), tumor necrosis factor-alpha (TNF-α) and nuclear factor-kappa beta (NF-κβ) and elevated the lowserum levels of total proteins and albumin.The high concentration of WGP and BGP ameliorated preneoplasticlesions induced by NDEA in liver tissue. The recent studies revealed that WGP and BGP counteracted NDEA-induced hepatic lipidperoxidation as evident by the increase of tissue reduced glutathioneand the decrease of malondialdehyde contents. White garlic powder and black garlic powder also alleviated NDEA-induced hepatic oxidative stress as manifested by restoration of activities of glutathione peroxidase (GPx), superoxide dismutase (SOD)and catalase (CAT)antioxidant enzymes. WGP and BGP antagonized NEDA-inducedelevation of8-OHdG content in liver DNA. In conclusion, these results revealed that both white and black garlic powders have good protective effect against NDEA-induced hepatocarcinoma. The study recommended that both white and black garlic powders should be beneficial for patients who suffer hepatocarcinoma إستهدف هذا البحث تقييم التأثير الواقى وفحص آليات العمل لتدعيم الغذاء بمسحوق الثوم الأبيض والأسود (المخمر) ضد ىسرطان الکبد الناجم عنن - نيتروزو داى ايثيل امين فى ذکور الفئران. فتم توزيع عدد42 من ذکور الفئران بطريقة عشوائية إلى 6 مجموعات کل منها 7 فئران کما يلى: المجموعة الأولى ضابطة سالبة (فئران غير مصابة) والمجموعة الثانية ضابطة موجبة (فئران مصابة بالسرطان)، والمجموعات الثالثة والرابعة والخامسة والسادسة فئران مصابة وسبق تغذيتها على وجبات مدعمة بمسحوق الثوم الأبيض والأسود المخمر وکلاهما بترکيز صغير( 2,5 %) وکبير (5 %) على التوالى. وتم إحداث الاصابة بحقن الفئران فى التجويف البريتونى بجرعة مفردة ( 100 مجم/کجم) من نيتروزو داى ايثيل امين فى الاسبوع الثالث من فترة التجربة ، وتلى ذلک حقن الفئران تحت الجلد برابع کلوريد الکربون(مخفف مع زيت البرافين بنسبة 1:1) بکمية2سم/کجم يوميا وذلک أثناء الأسابيع الرابع والخامس والسادس ( ثلاثة مرات فى الاسبوع) من فترة التجربة. وفى نهاية فترة التجربة تم تجميع عينات دم لفصل المصل وإستخدامه فى التحليلات البيوکيميائية، وتم أخذ نصف الأکباد لقياس دلائل أکسدة الدهون ونشاط اﻹنزيمات المضادة للأکسدة (سوبر أوکسيد دثميوتيز ٬ جلوتاثيون بيرأوکسيديز، کاتاليز) وترکيز مرکب 8- هيدروکسى- دى اوکسى جوانوزين (دليل موت الخلايا المبرمج ) فى نسيج الکبد.وإستخدم النصف الآخرﻹجراء الفحص الهستوباثولوجى للکبد. وأظهرت النتائج أن دعم الغذاء بمسحوق الثوم الأبيض والأسود المخمر قدأدى إلى نقص معنوى فى مستوى إنزيمات الکبد المرتفعة (اسبرتات أمينو ترانسفيريز٬ الانين أمينو ترانسفيريز والکالين فوسفاتيز) والبليروبين الکلى فى المصل٬ بينما أدى إلى زيادة بروتينات الدم. وأدى ايضا إلى نقص معنوى فى أدلة سرطان الکبد بالمصل بينما ادى الى زيادة نشاط اﻹنزيمات المضادة للأکسدة فى نسيج الکبد ونقص ترکيز مرکب 8- هيدروکسى - دى أوکسى جوانوزين. وأظهر الفحص الهستوباثولوجى وجود تحسن ملحوظ وإختفاء التغيرات المرضية المحدثة بمرکب نيتروزو داى ايثيل امين فى الکبد. ودلت النتائج على أن مسحوق الثوم الأبيض والأسود المخمر لهما تأثر واقى ضد سرطان للکبد. واشارت النتائج الى أن آليات عمل مسحوق الثوم کمضاد للسرطان عن طريق منعاکسدةالدهون بنسيج الکبد، وزيادة نشاط الانزيمات المضادة للأکسدة واحداث موت مبرمج فى الخلايا الکبدية المسرطنة. وتؤکد هذه النتائج على الاستخدام التقليدى للثوم فى الطب الشعبى للوقاية من سرطان الکبد. ولذلک توصى الدراسة أن تناول فصوص الثوم خاصة الأسود فى الغذاء مفيدا للمرضى الذين يعانون من سرطان الکبد نتيجة اﻹجهاد التأکسدى. وأن عزل المکونات النشطة بيولوجيا من الثوم ضرورى للبحث عن مواد طبيعية آمنة ﻹستخدامها فى الوقاية من السرطان بدلا من الأدوية الکيميائية التى يصاحبها غالبا أعراض جانية ضارة . | ||||
